Beschreibung

The lab of Peter Dirks investigates how medulloblastoma stem cells transition from a dormant state to an active, proliferative state, a process regulated by the transcription factor Olig2. The scientists found that inactivating Olig2 prevents stem cell activation, thereby blocking tumour growth. Targeting Olig2 with a small molecule inhibitor, CT-179, significantly slowed early tumour formation and regrowth after therapy in a mouse model. Further analysis revealed the drug effectively limits the ability of the quiescent stem cells to transition to a proliferative state. The study suggests that therapies targeting this stem cell fate transition could improve medulloblastoma treatment outcomes, especially in high-risk cases. The findings provide a basis for developing new treatments targeting cancer stem cell activation and limiting patient tumour progression. Research funding for pediatric brain tumours is at an all-time low.
