episode Tesamorelin: The Age-less Pulse of Growth Hormone | Age-Less Top 20 Peptides cover

Tesamorelin: The Age-less Pulse of Growth Hormone | Age-Less Top 20 Peptides

In the mid-1990s, antiretroviral therapy was transforming HIV from a death sentence into a chronic condition — and creating an unexpected syndrome in its wake. Patients on the new combinations were developing visceral fat accumulation, peripheral fat loss, dyslipidaemia, and a measurably blunted growth hormone rhythm. A small Canadian biotech in Montréal saw the lever. Native GHRH had a half-life of seven minutes, cleaved almost instantly by an enzyme called DPP-IV. Their structural innovation — adding a single trans-3-hexenoic acid group to the N-terminus — blocked the cleavage site, extended the half-life to twenty-six minutes, and made an entire pharmacology clinically viable. That compound is Tesamorelin, and in 2010 it became one of the very few synthetic peptides ever to clear the full FDA approval pathway. In this episode, Dr. Ethan walks through the molecule properly. The forty-four amino acid sequence, the structural modification that makes it work, and the mechanism that distinguishes it from every other growth hormone strategy: Tesamorelin doesn't introduce GH, it activates the body's own pituitary to release GH in its natural pulsatile pattern, preserving somatostatin feedback and the physiological rhythm that exogenous rhGH abolishes. The episode follows the cascade from GHRH receptor binding through cAMP and PKA activation to pulsatile GH release, hepatic IGF-1 production, and the anabolic and lipolytic effects downstream. Then the human evidence — and this is where Tesamorelin sits in a category of its own. Two pivotal Phase III trials, fifteen years of post-marketing data, and a defined safety profile in HIV-associated lipodystrophy. Emerging Phase II data in MASLD showing meaningful reductions in hepatic fat. Exploratory work in cognitive function, sarcopenia, and healthy ageing — promising mechanistically, but not yet pivotal-trial grade. Dr. Ethan closes with an honest 2026 assessment: what the off-label use looks like, what the safety considerations actually are, what diagnostics any responsible Tesamorelin protocol requires, and where the gap remains between mechanistic plausibility and demonstrated efficacy outside the approved population. As ever — what we know, what we don't, and what the honest picture looks like for anyone navigating this field. The Science of Age-less Living. Hosted by Dr. Ethan Hausman-Marquis. Can Thymosin Alpha-1 Keep the Immune System Age-less? | Age-Less Top 20 Peptides

In this episode of The Science of Age-less Living, Dr. Ethan explores Thymosin Alpha-1, one of the most clinically studied and widely prescribed peptides in the world. Originally isolated from the thymus gland in the 1970s, Thymosin Alpha-1 has evolved from an obscure immune peptide into a prescription immunomodulator used in more than 35 countries. We examine the remarkable history of its discovery, the molecular mechanisms that drive its effects, and the evidence supporting its use in chronic viral infections, sepsis, oncology, vaccine enhancement, and age-related immune decline. You'll learn how Thymosin Alpha-1 influences Toll-like receptors, dendritic cells, T-cells, and natural killer cells, and why many researchers consider it one of the most biologically interesting immune-regulating peptides available today. Most importantly, we separate evidence from speculation. Does Thymosin Alpha-1 genuinely support healthy ageing? Can it help counter immunosenescence? What do decades of clinical data tell us, and where do the limits of the science remain? No hype. No marketing. Just a deep dive into the biochemistry, clinical evidence, safety profile, and real-world applications of one of the most important peptides in modern immunology.

episode Klotho: The Longevity Hormone You've Never Heard Of | Age-Less Top 20 Peptides cover

Klotho: The Longevity Hormone You've Never Heard Of | Age-Less Top 20 Peptides

Klotho was discovered in 1997 when a disrupted gene caused mice to develop every hallmark of premature ageing within weeks — osteoporosis, arteriosclerosis, muscle wasting, cognitive decline — and die at a fraction of their expected lifespan. The inverse experiment extended lifespan by twenty to thirty percent. The protein was named after Clotho, the Greek Fate who spins the thread of life. It remains one of the most studied proteins in longevity biology nearly three decades later. In this episode, Dr. Ethan Hausman-Marquis examines Klotho from the ground up: the critical distinction between transmembrane α-Klotho and its circulating shed form sKlotho; how the FGF23 co-receptor mechanism explains the accelerated ageing phenotype of Klotho deficiency; what five separate longevity-associated pathways — including IGF-1/PI3K suppression, Wnt/β-catenin inhibition, Nrf2 antioxidant activation, and TGF-beta fibrosis suppression — suggest about why this protein matters so broadly; and why circulating Klotho levels decline by approximately 40 percent between the ages of 40 and 80. The human evidence is examined honestly: what the large prospective cohort studies actually show, what the KL-VS genetic variant data adds to the causal argument, what the 2014 Cell Reports cognitive findings mean for neurodegeneration research, and why the interventional human data — despite the extraordinary preclinical picture — is essentially non-existent. The evidence base, the interventional frontier, the exercise and vitamin D data, and the honest assessment of where exogenous Klotho research actually stands in 2026 — all of it, examined without hype. If you're researching Klotho supplementation, Klotho and ageing, Klotho and cognitive decline, or the science behind longevity peptides and anti-ageing medicine, this is the rigorous account the subject deserves. episode BPC-157: The Body's Own Repair Peptide | Age-Less Top 20 Peptides cover

BPC-157: The Body's Own Repair Peptide | Age-Less Top 20 Peptides

This is the first episode in a new series: the Top 20 Peptides. Over twenty episodes, Dr. Ethan Hausman-Marquis examines the most researched compounds in longevity and regenerative medicine — the real biochemistry, the honest evidence base, and the regulatory context behind each one. No hype, no discount codes. Just the science. We start with BPC-157. Derived from a naturally occurring sequence in human gastric juice and studied for over thirty years, it's the peptide that put this whole field on the map — and one of the most misunderstood molecules in it. This episode covers how it was discovered, what its proposed mechanisms actually are at the molecular level, what the preclinical research genuinely shows across five areas of investigation, and why a 2025 systematic review of 544 published articles found exactly one human clinical study meeting inclusion criteria. The evidence base, the open safety questions, the FDA compounding restriction, the WADA prohibition — all of it, examined honestly. If you're new to peptide science, this series is the foundation. If you're already in the space, this is the rigorous re-examination it deserves. The Age-Less Top 20 Peptides series. Episode 1 of 20.

episode Stack Smart: Supplements + Peptide Therapy for True Age-less Living cover

Stack Smart: Supplements + Peptide Therapy for True Age-less Living

In this episode of The Science of Age-less Living, Dr. Ethan Hausman-Marquis reveals why many people using peptide therapy only achieve partial results — and how an easy, science-backed fix can unlock the missing 30–40% of their potential. He explains that peptides (such as BPC-157, CJC-1295/Ipamorelin, Thymosin Alpha-1, and collagen peptides) are powerful signaling molecules that tell your body to heal, regenerate, reduce inflammation, and optimize hormones. However, they cannot create results out of nothing. They require specific nutrient co-factors, low inflammation, and proper cellular conditions to deliver their full effects. He breaks down the key synergies between peptides and supplements, highlighting four powerful “Power Pairings”: * BPC-157 + Zinc & Magnesium for superior gut healing and tissue repair * CJC-1295/Ipamorelin + Vitamin D & Zinc for maximized growth hormone response * Collagen Peptides + Vitamin C for better skin, joints, and connective tissue * Thymosin Alpha-1 + Vitamins C, D & Selenium for stronger immune function The episode also covers the most common deficiencies (magnesium, Vitamin D, zinc, and omega-3s) that quietly sabotage peptide results, the importance of blood testing, and practical baseline stacks. Dr. Ethan shares how The Catalyst Clinic uses comprehensive testing, personalized analysis through their AI system AURION™, and custom-compounded daily supplement packets to make optimal stacking effortless and highly effective. Key Takeaway: Peptides signal. Supplements supply. Stack them intelligently, and you’ll move from good results to truly transformative Age-less Living.
